How to Reconstitute Research Peptides: A Step-by-Step Guide

Reconstitution is the process of dissolving a lyophilized (freeze-dried) peptide powder into a liquid solution for use in research. Done correctly, it preserves the integrity of the compound and ensures consistent results across experiments. Here’s what every researcher should know.

Choosing Your Solvent

Bacteriostatic water (sterile water containing 0.9% benzyl alcohol) is the most commonly used solvent for peptide reconstitution in research settings. It inhibits microbial growth and extends the usable life of the solution. For peptides that are difficult to dissolve, a small amount of dilute acetic acid (0.1%) can be used as an initial solvent before diluting to volume with bacteriostatic water.

Step-by-Step Reconstitution

  1. Wipe the rubber stopper of the peptide vial and solvent vial with an alcohol swab and allow to air dry.
  2. Draw the desired volume of bacteriostatic water into the syringe.
  3. Insert the needle into the peptide vial at an angle, directing the solvent toward the glass wall — never directly onto the powder, as this can cause degradation.
  4. Gently swirl (do not shake) until the powder is fully dissolved. The solution should be clear.
  5. Label the vial with the peptide name, concentration, date of reconstitution, and your initials.

Storage After Reconstitution

Reconstituted peptides should be stored refrigerated (2–8°C) and protected from light. Most reconstituted solutions remain stable for several weeks under proper refrigeration. For long-term storage, lyophilized peptides should remain frozen at -20°C or below until ready for use.
